The Alfa Laval SB Carlsberg Flask consists of a cylindrical container with a flat bottom and top cover. It is equipped with breathing filters, a membrane sample valve for aeration and product transfer, and a micro sample port that allows aseptic introduction of pure yeast culture by syringe.
For operation, sterilized wort is filled into the flask to approximately 80% of its total volume. Sterilization can be carried out using an autoclave, gas burner, or electric hotplate. After sterilization, the flask is cooled in a refrigerator or cold room to reach the proper working temperature.
The cold wort is then aerated through the membrane sample valve using an aeration lance. Sterile air is required during this stage to maintain hygienic conditions. Adequate air supply is important to reduce foaming and to prevent the breathing filter from being exposed to foam.
Once the wort has been sufficiently oxygenated, the yeast culture can be introduced aseptically through the membrane fitting using a syringe. Dry yeast culture may also be transferred through the empty filter housing. This process allows the Carlsberg Flask to support controlled early-stage yeast propagation under sanitary conditions.
At the appropriate growth stage, the yeast culture can be transferred from the flask to the inoculator or yeast propagation plant. Before transfer, all connections, hoses, and valves should be disinfected to maintain aseptic handling. After use, the flask should be disassembled and cleaned by hand using standard detergents.
